I'm in Great Britain and would generally use British punctuation and spelling (e.g. single quotes, and quotes inside end punctuation where the quoted material isn't a whole sentence). Will I be marked down? If so, I'll change it.

This would be fine, as we usually advise judges not to criticize any non-standard spelling or punctuation. But we do ask that the assigned word be included exactly as it is spelled. Please let us know if you have any other questions or need anything else. Good luck!

There is an F.A.Q. at the top of this forum section which states:
I use Canadian spelling (much closer to British than American), and I've not had a judge comment on it so far. I would hope all judges are familiar with at least the most common differences.
